The rat hepatoma cell line McA-RH7777 was transfected with a DNA encoding a Fas-ligand fusion protein. Fas-ligand protein expression was not toxic to the cells, but induced apoptosis in a concentration-dependent fashion. Apoptosis was detected by DNA laddering, phosphatidylserine exposure, and cell shrinkage. We also analyzed the expression of Fas by flow cytometry and Western blot analysis. The intensity of staining for Fas was dependent on the concentration of Fas-ligand protein. Treatment of the transfected cells with anti-Fas antibody and with TRAIL induced apoptosis. These results suggest that Fas expression can be used as a predictive indicator for the effectiveness of Fas-ligand-induced apoptosis.Q:
